Bis-Tris Gels Technical Information

RunBlue® Bis-Tris Protein Gels have been developed by Expedeon™ to provide an alternative to NuPAGE® Bis-Tris gels. These protein gels run with the same voltages, in the same tanks, using the same MES/MOPS buffers as NuPAGE gels. RunBlue Bis-Tris gels achieve highly comparable results with exceptional reproducibility and resolution.

Polyacrylamide gels are based on the polymerisation of liquid acrylamide and bis-acrylamide. The polymerisation is usually induced by the addition of a source of free radicals and a stabilizer (usually APS & TEMED) which induces cross link formation between acrylamide and bis-acrylamide monomers. This localised chemical induction results in heterogeneous polymerisation across the gel due to acrylamide concentration gradients being generated, and can lead to the presence of residual free acrylamide at the end of the polymerisation.

Expedeon has developed a unique physical induction technology which ensures homogenous polymerisation across the whole gel and no residual free acrylamide after the end of the polymerisation. RunBlue gels are polymerised in this unique homogenous manner, resulting in a consistent gel with minimal or no residue after casting.


Don’t change a thing

Expedeon’s new Bis-Tris gels can be run with standard MES or MOPS buffers depending on the molecular weight range of proteins to be separated and use the same LDS sample buffer and protocols as NuPAGE (200V). The cassettes fit in the NuPAGE SureLock™ tank without any need for adaptors or spacers, and gels are also compatible with the iBlot® dry blotting system. There is no workflow disruption, no downtime, and no transition period.


Don’t compromise on results

Expedeon have over 8 years’ experience in pre-cast gel manufacture and have applied this knowledge to Bis-Tris gel chemistry. A unique manufacturing process with exacting specifications and quality controls results in increased batch to batch consistency between gels. A key element in this process is the use of physically induced gel polymerisation for consistency, as oppose to chemically induced methods which can introduce variability leading to heterogeneous polymerisation. Switching to RunBlue Bis-Tris gels will give better resolution while following the same migration profiles.


Get added benefits

The RunBlue gel range has a number of usability enhancements. A unique composition gives strength with a slight elastic stretch, greatly reducing the risk of tearing or damaging gels. The comb is removed during the manufacturing process to eliminate the risk of damaged or broken gel teeth, and there is no strip to remove from the cassette. Extra wide and deep wells allow loading of samples up to 35 µl in volume for a 12 well gel. A 2 year shelf life at 4°C or 3 months at room temperature provides flexible storage options.
