Ponceau S Solution
Our Ponceau S solution is a staining reagent for the detection of proteins on nitrocellulose and PVDF membranes. Ponceau S solution allows users to check the efficiency of protein transfer to blotting membranes during Western blotting before membrane blocking and protein detection.

Users can directly incubate the blotting membrane (PVDF or nitrocellulose) in ~15ml of Ponceau S solution following protein transfer. The transferred protein becomes rapidly visible as red bands on the membrane, with the highest intensity of staining reached within five minutes of incubation with Ponceau S solution. Rinsing the membrane with water after Ponceau S staining can help reduce ‘background’ staining on the blotting membrane. The intensity of staining will correspond to the amount of protein transferred.

Is Ponceau S solution suitable for both nitrocellulose and PVDF membranes?
Yes, it is. Ponceau S solution has been tested on both nitrocellulose and PVDF membranes and it provided reproducible data as well as a high signal-to-noise ratio.
Is Ponceau S solution suitable with different blocking agents other than BSA?
Yes, it is. Ponceau S solution is suitable with the most common-use blocking agents, such as non-fat dry milk (3-5%), casein (1%) and fish skin gelatin (1%).
What is the recommended dilution for Ponceau S solution ?

Can Ponceau S solution be used to detect proteins under both reducing and non-reducing conditions?
Yes, it can. Ponceau S solution has been tested on proteins run under both reducing and non-reducing conditions, with both conditions showing equal sensitivity of detection and reproducibility.
What is the limit of detection for Ponceau S solution?
Ponceau S solution can stain as little as 250ng of Bovine Serum Albumin on a nitrocellulose membrane.
